Greetings, We have run about 2 dozen CLL cases against ZAP-70 from various vendors. Gating on T cells from a normal blood shows good positivity with antibody from all vendors. The CLL lymphocytes have been either negative or dimly positive compared to T cells, however. I haven't been able to find any clearly positive cases. This runs counter to the idea that ZAP-70 expression works as a surrogate marker for unmutated IgVH gene status, which occurs in roughly half of CLL patients. I will append a couple of histograms showing a case with very slight B-cell positivity, and would appreciate any technical suggestions. We are running on a Beckman Coulter XL, staining cells after permiabilization with Coulter IntraPrep reagent. Is anyone getting unambiguous results with ZAP-70 testing by flow?
