small things

Hello Group,

I am trying to sort bacteria on a MoFlo with a 70 micron tip at 60psi.	My
problem is that when triggering on FS or SSC a large percentage of my
cells fall within the "noise".	If the bugs are labeled with a bright
fluorophore, I can trigger on a fluorescent parameter with no problem.
Since I need to keep FL1 free as my sorting parameter, I'd like to label
all the bacteria with a dye that has adequate excitation at 488 and
emission (red) that doesn't fall into my 530 FL1 filter. Does anyone in
flow-land know of a dye that will label all (or at least viable) bacteria,
has red emission, and is non-toxic to bacteria?

Also, for you jet-in-air sorters.... what is the lower size limit in your
hands of particle size that is distinguishable from noise on FS and SSC? I
can see 0.9 micron beads fine triggering on FS, but the bacteria are more
problematic.

Thanks very much,
