Flow count flourosheres

Dear All,
 
We want to quantify the amount of endothelial cells (big cells > 10 um)
in whole blood by using Flow count flourosheres beads. Does somebody
have experience with these kind of experiments. We did some preliminary
experiment in which we saw that the detection was linear had high amount
of cells, but at low amount of cells it seems to me that the small
particles of the Flow count flourosheres beads are causing troubles (we
have to count more then 2 million cells to get enough endothelial cells
(+/- 200 to analyse). Do I have to use bigger beads or more beads.
