Intracellular Cytokine Staining

Hi Fellow Flowers,
 
We are applying the traditional protocols for the stimulation and detection
of Cytokine and are experiencing problems with the detection of IL-10, which
is not appearing.

As we all know, the results obtained using frozen samples are different. We
have tested this by using the same linfos taken from the same patient.

The results show:

When fresh linfos are used, IL-10 does appear. However, when we use frozen
Lymphocytes, there is no appearance of IL-10.

My question is: should we use an anti CD3 to increase the production of
IL-10?

I would be very grateful if someone could help me with this query.
