Hi, FLOWers, we started using SpectrAlign (purchased from DakoCytomation, cat # K0111) for instrument alignment. The advantage of the beads is that they get excited by every lasers we have and emit light in very wide spectrum. We were happy with it for a few weeks until I noticed the Fl-3 channel intensity decrease on a Calibur. Only that particular detector. I moved to the other Calibur, recalled the alignment page and setting. Same observation. Fl-3 dropped by the same rate. The Fl-3 is a 670 LP on the Calibur. There is no loss in Fl-4. The beads were stored cold and dark. The dilution was made freshly before use in PBS. Are the beads losing high wavelength intensity? Has anyone experienced the same? Best, Akos
