Hi,
The viability of sorting cell is a general question but especially on
high-speed sorter. For evaluation of viability, you can use annexin V with
7AAD, but I prefer to use the CSFE to view the proliferation of sorting
cells because I have examples of cells that are alive after the sorting but
doesn’t proliferate.
So using the CSFE protocol, you can evaluate, for the different kind of cell
you sort, the conditions of pressure (limit of pressure) and speed (limit of
speed). For a given pressure, with certain cell, you can have a limit of
speed.
