Dear Flow-ers, I was wondering if anyone has been using TO-PRO-3 to measure DNA content on a 4 color FACS Calibur. We are using FITC, PE, & Tri-color labeled antibodies, against surface antigens and intracellular cytokines (thus including saponin permeabilization). Therefore, we are trying to measure DNA content simultaneously using FL4 to detect TO-PRO-3 binding. I was wondering if other investigators have had success using such an approach.
