Fellow Flowlanders, We have a customer that is interested in using a lacZ B-Galactosidase detection kit on our LSRII, which is currently equipped with the 325nm HeCd laser. The information we have found indicates that it is optimally excited around 390nm but we have been unable to locate a spectral graph of the excitation of this substrate. Does anyone have experience exciting CUG (3-carboxyumbelliferyI B-D-galactopyranoside) with this laser line? We would appreciate any suggestions or hints that you can offer.
