Dear flowers,

i am trying to standardize phagocytosis assay using FITC labeled heat
killed E.coli in case of peritoneal macrophages...
i used crystal violet (0.2% stock soln. final conc. 0.01%) to quench the
fluorescence of unphagocytosed bugs....i am facing following problems:
1. some times i get clean peak shifts...some times don't...
2. i have found that at higher bug numebrs crystal violet fails to quench
the background.....
3. how the typical positive shift will look like?...
