Hello. I've been extremely frustrated with the Annexin V/PI kit from Molecular Probes, and wonder what others' experience with it has been, specifically using it with flow. Also, if there are similar kits from other vendors that someone has had good luck with, that would also be helpful. If PI fluorescence is graphed on the y axis and annexin on the x, I seem to get very low annexin signal compared to the PI; even with compensation, I can't avoid having a large population of cells showing up in the upper left quadrant (PI only) which should be empty, because as I understand it cells "leaky" to PI would also bind the annexin.
