AlexaFluor conjugations

Hi all,
We are looking for some feedback on AlexaFluor antibody
conjugations. We have been using the kits from Molecular Probes
("Protein Labelling Kits") and have been getting mixed results.

We are wondering what experience people have had with the kits - what
factors might affect the reproducibility? (individual antibodies? age
of the kit? etc)

We have also been thinking about using just the straight succinimidyl
esters (for cost reasons and also so we can experiment with the ratio
of dye to antibody). However, we are a little concerned about the
statement in the product information that "when stored properly,
these reactive dyes should be stable for at least 3 months". We
understand the need for Molecular Probes to be clear and conservative
about how long they expect the desiccated dyes to last but we are
wondering what is general experience with these dyes? Will they be
unusable after 3 months? 6 months? 12 months? We will probably be
doing a number of conjugations with each AlexaFluor but I doubt we do
them all within 3 months...

Thanks,

Adrian Smith
