The sub G1 peak you may visualized does not differentiate between apoptosis and necrosis; therefore an Annexin V type assay would help to confirm your result. Also I will not exclude the bottom debris region as this is where you will see a sub G1 peak. You may also try analyze your data with Modfit 3.0 from Verity software House. The PI fluroscence is being acquired in the log scale. FSC and SSC are in the linear. Right now i am unable to do the dual staining with Annexin. But i would like to know if it is fine if i produce my results only with the PI staining. Also i am not sure if the bottom debris region at the FSC/SSC should be included or not during the analysis.
