Hello!! I am a cardiovascular investigator trying to isolate vascular smooth muscle cells from rat carotid arteries at as high purity as possible. Unfortunately, a previous attempt showed that only 25% of isolated cells were alpha-actin positive after differentiation (heparin and 0.5% sera for 4 days). Other cells appear to be endothelial cells. Does anybody have a good protocol for vascular smooth muscle cell isolation?? In addition, do you have any cleaver way to use FACS to determine the purity of the prep? I thank you for your time and help on this. Sincerely, Ken
