I do immunocytochemistry for a cytoplasmic antigen and the count at the microscope was very low (0.03%). When I do the same reaction evaluated whith FACScalibur the result was 0%. However when the immunocytochemistry was 0.3%, the FACS evaluated 2%. This evaluation could be correct? May depend from instrumental chracteristic rhater than error in protocol because the percentage was low? In general the count whith FACS is accurate and I can appreciate 0.02 or 0.2% of cell staining?
