Dear all, An investigator in the lab try to stain sperm cells in order to differentiate spermatozoid, aploid germinal cells and diploid non germinal cells. First we tried to stain simultaneously spermatozoid and lymphocytes using PI (Vindelov staining technic) and found a 4.6 fluorescence intensity ratio between the two populations. Can DNA structure (compaction, protamines vs histones) explain this observation? Any comments will be welcome. Thanks in advance. Franck.
