Hi Maryalice, I used to put the tissue in tripsin-edta for 30-60 minutes at 37 C, and after that is easy to separate epidermis from dermis. We get good recovering of Dendritic cells. You can also use dispase or DNase in order to get the full cell suspensions. I hope it will help, regards Rafael >Has any one gotten good cell recovery with surface antigens intact >from skin biopsies? The cells are adult T cell leukemia/lymphoma >cells and are a little fragile. We tried mincing and just got debris. >We are thinking of aspirating the lesion. Any bright ideas? > > > Maryalice >Maryalice Stetler-Stevenson >Director Flow Cytometry Unit >Laboratory of Pathology, NCI, NIH \|/ (o o) ________________________________oOo__(_)__oOo_________________________________ ___/\_ | Rafael Nunez mailto:rafaeln@vetvir.unizh.ch / o \/| | University Inst.for Virology http://www.vetvir.unizh.ch/ / _| | Winterthurerstr. 266a Telephone: (+41) 1 6358710 /_/\__/-\/ | 8057 Zurich SWITZERLAND Faximile : (+41) 1 6358911 ______________________________________________________________________________Received on Fri Feb 25 12:32:57 2000
