I'm considering Dynal and Advanced Magnetics > antiRabbit IGg products, but have heard Dynal beads give low recovery > rates. I've done similar things, though not with filamentous bacteria. I'm puzzled as to why Dynal beads are reputed to give low recovery rates. The limiting factor as far as I could tell was the hapten and the labelling step. Recovering the beads was rarely a problem, even from blended soil samples. Linking the hapten onto the beads whilst maintaining reactivity was more of a problem, but it was straightforward to improve this by reacting biotinylated hapten with the cells and then adding streptavidin coated beads.
