Hi Cindy We use CD138/CD38/CD45. Plasma cells are CD138+CD38++ and usually (but not necessarily) CD45-. If you set up a region on the CD138+/CD38++ and backgate to a FSC/SSC plot the cells appear to the right of monocytes, (i.e. larger, and sometimes more granular), and if there's not a huge and distinctive population I find it easier to look for them on this plot than to wonder whether they might or might not be weakly CD45+ and disappear into the neutrophils on a CD45/SSC plot.
