Hi, Slava! I am currently working on a project that uses flow cytometry to measure the in vivo production of intracellular IFNg and IL-2 by cytotoxic T lymphocytes in the murine P815 allogeneic model. As for measuring cytokine production by ELISA techniques, I cannot detect these cytokines straight out of the animal. However, I can detect these cytokines following ex vivo restimulation of the spleen cells for 4-6 hours in vitro with P815 tumor cells. In this way, I can examine the production of IFNg and IL-2 by antigen-specific effector cells. I can supply you with further details if interested.
