Is there anyone who has used the subtraction histogram function on
CellQuest? How useful is this
quasi-histogram? Is there an appropriate time when finding the difference
between two histograms this way
is valid? And then what about overlaying these histograms with others?
Any comments? It seems reasonable
to use this function when comparing {isotype and/or unstained samples-test
sample pairs} with other
{isotype and/or unstained- test samples}. Would it provide you with a sum
histogram, that when compared with other sets, compensate for the slight
differences in autofluorescence between sets? Or is the subtraction
histogram
a fancy software component of little use?
