Dear flowers, I'm preparing to measure the expression of CD40L on human T-lymphocytes (from peripheral blood and metastatic lymph nodes) upon stimulation. 1) What stimulating agent is the most appropriate to use (is it possible to induce it with stimulators such as PHA or IL-2 or just with PMA and Ionomycin), because I didn't find any induction of CD40L after 18 hours with IL-2 (200 IU/ML), neither with anti-CD2\CD2R (B-D). 2) What would be the best time following stimulation to analyse it on surface? 3) Is it possible to do that in cytoplasm? (rather not because it's PE) Oh yes, anti CD40L PE (or CD154) came from B-D.
