DMSO as a solvent

Hi all in FlowLand--

I have been using rhodamine-123 as an indicator of the mitochondrial
membrane potential in human fibroblasts for quite some time with good
results. Currently, my stock is dissolved in double distilled water.
However, lately I have had to run some experiments to test the effects of
DMSO on the rhodamine fluorescence in these cells, and I am finding that the
DMSO is increasing the Rh-123 fluorescence by 20-25%. Is this a normal
phenomenon? I am posing this question because some other dyes, namely
Fluo-4 and Rhod-2, are dissolved in DMSO and I am concerned that this
solvent is skewing my results (which is important because I have my master's
thesis on the line!).

Speaking of fluo-4, have any of you had experience with it? I am obtaining
very broad fluorescence peaks when I use it. Is that normal?

Thanks in advance for any information!!
