Dear Dr. Lin,
In response to your questions regarding platelet function testing in
patients undergoing cardiac bypass surgery, a number of studies (flow
and non) have looked at platelet and coagulation parameters during
bypass. Our own experiance (see Ann Thorac. Surg. 61:1188, 1996)
indicates that: 1) aprotinin maintains platelets unstimulated during
bypass, and maintains the von-Willebrand pgIb mediated platelet
activation pathway; and, 2) analysis of platelet-WBC aggregates are
more revealing than platelet-platelet aggregates. Regarding the LIBS
issue, most of the antibodies available thus far appear to be
sensitive to fixation, making activation related studies a bit
difficult (ie, activating the sample with an agonist to determine the
platelet "activatability"). For some information regarding LIBS (and
RIBS), please see the European Working Group Consensus on Platelet
Flow Cytometry (Thromb. Haemost. 79:885, 1998).
