Dear Sonja We use DAPI on fixed cells at approx. 2ug/ml adding 20ul per test and allowing them to stain overnight. Fresh cells need a permeabilization stage (0.2% Triton X-100 in PBS for example. Add 100ul and mix for one minute) then add DAPI and run within 20 minutes. Fresh cell staining is very rapid. Best wishes, John.
