Nancy: Some time ago, we looked at the effect of storing fixed, labeled platelets for different time periods, at 4 deg. C. The bottom line is that for whole blood analysis, the platelet markers are generally equivalent, comparing samples run after 24 hrs vs. immediate (ie. within 1-2 hrs) analysis. After 24hrs, the results get quite variable. Although we did not look at platelet rich plasma extensively, our impression was similar, though PRP samples were more variable after 24 hr storage at 4 deg. C. We rarely use PRP these days, so I can't comment beyond this. However, our rule for whole blood platelet analysis is to run soon after Ab incubation, if possible, or within 24 hr. if not.
