          The more recent Zynaxis PKH dyes (sold by Sigma) as well as 
          the carboxyfluorescein succinimidyl ester (CFS) (J.Cell 
          Biol.101, 610; ;1985 and 103,2649;1986) are both good 
          tracker dyes.  I have advertised the latter for a long time, 
          and use it with bacteria for that purpose. When using the 
          succinimidyl ester dyes the carboxy-dichloro-fluorescein 
          diacetate succinimidyl ester (CCFAS, #LA-572, Lamda 
          fluorescence technology, Graz) has an advantage of a lower 
          pKa (4.5) thus fluorescence is less pH dependent.  The 
          advantage of CCFAS is that initially only 'vital' cells are 
          labeled and the dye binds intracellular, but the dependence 
          of esterase activity also enhances variation and cell 
          volume.  The PKH dyes bind to all cell membranes and might 
          (have not compared it) therefore give smaller CV's.  Also 
          that cell wall is a square and cell volume the cube function 
          of variations in cell diameter might be of advantage. I was 
          always interested in a double labeling experiment of those 
          two cell tracker systems to see how daughter cells share 
          membranes and cytoplasm.  Perhaps one day...

Anyone know of a fluorescent marker/stain that would survive culturing
of cells for 1 to 2 days.  I would like to track these cells through
various depletion procedures.
