In a recent paper from Mattis AE et al, Journal of Immunological methods
204 (1997) 135-142, they use live cells to "analyzing cytotoxic T
lymphocyte activity: a simple and reliable flow cytometry-based assay" by
double staining with DiO18 (green florescent dye) and PI. The FACS data are
quite clean. May be their protocolls could help you.
