Yes, you can use FACS Assistant to calculate ratio, you can also export
your data as mean ratio against time and display using cricket graph or any
other graphing program. I use it all the time. If you promise to pay Ray
Hicks the exorbitant price of 25 bucks or whatever  he will send it to you.
