Hi all, My hospital has a contract with a specialty lab that reports the CD34+ yield from our stem cell harvests. I noticed that that lab doesn't perform progenitor assays (CFU-GM). I knew that things were headed that direction, but how many of you (or your ref. labs) have dropped the colony assay in favor of flow cytometric measurement? Also, for same-day prediction of the yield to be reported, I'll be running a few tubes on peripheral blood: ISHAGE method, CD45, CD34, and a viability dye. I'd like to add in CD38 or Thy-1 (limited funds) to define the less committed portion of the total CD34+ population. Will either one of those be useful (w/ CD34)? or am I missing out if I don't do both or if I don't prove the population to be negative for lineage markers as well? Thanks for any input.
