The problem with supravital cell staining with pyronine Y is that at
relatively low concentration (below 5 uM) the dye, similar as rhodamine
123, is localized in mitochondria, photosensitizes cells and binds
minimally to RNA. At that concentration it arrests cells in G1. At
concentration above 5 uM it also binds to RNA and then arrests cells in
S (Cancer Res., 46: 5760, 1986). Pyronine Y can be used as a probe of
RNA after cell fixation (Cytometry, 8: 138, 1987).
