Hi, I have just joined the list. I'm particularly interested in knowing other workers interested in flow cytometry of plant material (probably an enthusiastic minority!). I am currently using a Partec CA-III to determine DNA amounts in roses. This is a particularly difficult species viz. phenolics. Does anyone have experience with contolling phenolics in plant tissue as we have found they cause a great deal of fluorescence quenching, at least with DAPI staining. I am also interested to know if Hoechst dyes have a different permeability to intact plant cells than does DAPI. Thanks
