Need help with staining of human neutrophils for fluorescent microscopy
(cytoskeletal proteins).My protocol works fine for platelets and lymphocytes,
but not for neutrophils. After fixing with 2% paraformaldehyde and
permeabilizing with 0.2-05 % Triton cells are negative even for actin. I tried
another protocol: fixing with 1% glutaraldehyde, permeb. with 0.1% NP40 -
stains fine for actin, but negative control is as bright as specific staining.
