Steve I routinely use 2 750 instruments for 3 and 4 color applications. There are a couple of solutions for your compensation woes. A hardware approach is to acquire a "subtraction module" from an old EPICS V system. This will plug right into the lower rack of your 750 and give you another compensation channel. This has the benefit of allowing you to correct the 3rd color (Pe-Cy5) using a PE signal already corrected for the FITC overlap. Alternatively Cytomation has done a pretty good job with adding compensation to their system. You can compensate the 3rd channel there and use it for sorting. We have Ciceros on our systems. gary Gary Durack University of Illinois at Urbana-Champaign Biotechnology Center Flow Cytometry Facility 217 244 0559
