We have been routinely running 4 colour analysis on HIV patients on our XL with FITC, PE, ECD and PerCP, although not with the combinations you suggest. It works very well, with careful compensation, although it can depend somewhat on the "brightness" of your antibodies. Compensating PE out of ECD and vice versa seem to be the worst. We are usually using CD3 PerCP, CD4 or CD8 ECD and using PE and FITC for other markers of interest.
