I remember when he edited a German book on Clinical Methods
in Flow Cytometry (subsequently published by Schmitz et al)
that they had a routine protocol for heparinised whole blood
          called:

          Detection of intracellular cytokine
          - functional characterisation of immunecompetent cells in
          immune defects
          - monitoring of immunosuppressive therapy

from a group in Bochum. They used a system of isotype
specific secondary antibodies and triple colour
          fluorescence.

Two questions about detection of intracellular cytokines:

1) Does anyone have an opinion concerning the relative merits of
Brefeldin A and monensin for blocking cytokine secretion?

2) Does anyone have information on the availability (commercial or
otherwise) of directly conjugated anti-cytokine antibodies. We have
only been able to come up with one (FITC anti-IL2 from Biosource).

Thanks,
