Mikael, Dr. Martz is correct, the FCS list mode files produced by version 1.0 of CELLQuest do not contain instrument settings, I am sorry if this has caused you any problems. Version 1.1.1, which is now shipping to FACSCalibur customers, does write a complete set of instrument settings to the list mode file. The instrument settings can now be recalled and downloaded from either a list mode file or a separate instrument settings file. An update for your 1.0 version of CELLQuest will be available in the near future. Here is an excerpt from the on-line help of my free program MFI: [BEGIN QUOTE] CYTOMETER/SOFTWARE FILES TESTED [...] FACStation 1.0 'CellQuest' MacIntosh System Software 7.1.0 (FACScan) *** [...] *** The FACStation 1.0/MacIntosh files tested do not include instrument gains, PMV's, threshold, or compensation. Becton-Dickinson promises to rectify this serious deficiency in a future upgrade. This deficiency precludes MFI from detecting inadvertant changes in instrument settings. [END QUOTE] The above statement is based on my September, 1994 correspondence with Michael Cosgrove, B-D's project manager for CELLQuest. I have CC'd this message to Mr. Cosgrove -- perhaps he can let us know when the upgrade can be expected? /*- - - - - - - - - - - - - - - - - - - - - - - - - - - - - - - - - - Eric Martz, Professor of Immunology emartz@microbio.umass.edu Dept Microbiology Voice: 413-545-2325 FAX: 413-545-1578 Morrill IVN 203, Box 35720, Univ Massachusetts, Amherst MA 01003-5720 - - - - - - - - - - - - - - - - - - - - - - - - - - - - - - - - - - -*/
