Hello everyone We have problems with the MultiPlus Software from Phoenix Flow system. We had the MultiPlus Ver. 2.5 to analyse 2 dimensional DNA histograms from the BrdU/Hoechst/EB method. We get listmode files from the Coulter Elite ESP in FCS1 standart. We had the following problem: this version had problems with setting a gate before analysis. On the other hand, it was unstable after about 10 analysis. I complained about this problems by Mr. Becker from Phoenix and bought a new version, version 3.0. He told me, that this version should not have this mistakes. We tested this version last week and have still the problem, that the program is unstable after about 7 to 10 analysis. That means, after setting a region in the dot plot histgram and trying to analyse the DNA histogram - changing to MCycle - the program crash and one have to reboot the computer. I know, that we need a minimum of 560 k free memory in the lower 640 K (we use a PC). Thats no problem, as we have 584K free. Mr. Becker wrote us, that he analysed about 40 samples without any problem. I got the following result: When I set a gate - peak versus area of DNA fluorescence to eliminate clumps of cells - the program remains unstable. When I directly take the listmode files for analysis, we have no problem. The second problem of the new MPlus software is the annotate function. One can annotate the histograms, but it is impossible to print it. This is a specific problem of the new version. Has anybody the same problems with MultiPlus. Can anybody help?
