To Jim Henthorn: From a number of sources we know that formaldehyde or paraformaldehyde will inactivate HIV-infected cells. In my experience, the number of infected cells (and HIV copies/cell) in the cell preparation (infected cell line vs. whole blood from HIV-positive donor) determines how much fixative for how long. To completely inactivate a cell line with 100% positive cells at a high copy number, I recommend overnight fixation in 1% formaldehyde or paraformaldehyde in the refrigerator. For whole blood, 1/2 hour under the same conditions is probably O.K. The biggest issue with sorting cells comes when the cells are not fixed.
