Interesting (and annoying) problem. I have some questions

(1) On whole blood, did the RBC stain ? (I wasn't shure if I got that right)
(2) Does this happen with samples from different donors and/or with different
    TCR (alpha/beta) antibodies ?
(3) What happens if you stain isolated (enriched) RBC e.g. from a ficoll pellet
(4) I assume your antibody is directly labelled (e.g. FITC). Try doing a
double staining with CD3 (PE) on PBMC. You should get a diagonal staining
pattern if the specificity is right (for the alpha/beta). You could then
label isolated RBC the same way and see how they look like.
(5) Is the staining intensity of CD3 and your TCR mab similar on isolated PBL ?
(6) For further consideration could you please give details of the antibodies
    used?

Thanks in advance |

Martin
