Kristi R. Hawkins requested info concerning measurement of oxidative burst in
macrphages isolated by adherence.

Our experience with human monos would suggest that by the time you have
isolated macrophages by adherence they are so "activated" that they are not
capable of mounting an additional oxidative burst in response to an agonist. 
The response to PMA is by far the greatest response you will observe so I
doubt that any other agonist will give you a response if PMA does not.
Perhaps you could try identifying the macs in suspension, without adherence, if
appropriate antibodies are available (probably not) or you could try resting
the macs for several hours or overnight in the hopes that they will return to a
more basal state which will allow you see an increase after stimulation.
