Bead CV's vary considerably with manufacturer, manufacturing process, and lot number, among other things. Over the years, the beads I have found to consistently have the lowest CV's have been 2 micron green spheres from Polysciences, which come out around 1.2 % in scatter and fluorescence in the best systems on the best days. I've gotten 1.2 % in a Cytopup with arc lamp illumination at 436 nm, and in laser systems, including one with a 441 nm, 100 mW HeCd laser. If the lamp or laser noise is 1% RMS, the CV won't be any lower than that, in the absence of saturation and bleaching effects. Not all bead lots are that good. --Howard
