We are requesting help in the resolution of a technical problem. We are attempting to study cell cycle kinetics using nuclei extracted from formalin fixed-paraffin embedded rat liver tissue. The method works well and good quality histograms are generated. The problem is that the estimated S-phase in normal livers is 10-15 percent using B/D's RFIT software. In fresh, ethanol fixed liver tissue the S-phase is 1-2 percent. So the questions are: 1. What is the most likely reason for the disparity in the data ? 2. Is it appropriate to use the S-phase data from formalin fixed liver nuclei as a control value in spite of the fact that it is higher than reported values for fresh tissue? Thanks for your help.
