I have a investigator using Celector Flasks for CD-34 enrichment.
After following the protocol, the cells are stained for CD 34 and run on
the FACScan. To date we have not seen any staining greater than background.
Question? Is it possible that the antibody on the flask is
remaining on the cells and blocking any subsequent staining?
We would like to develop the cells with a second step
reagent, but to what? Is the anti-CD 34 a mouse? rat? rabbit etc?
There has been no response from the company to date.
Have not used the CD34 AIS flasks but we use the CD4 and CD8 flasks. After allowing the cells to spontaneously detach from the flasks (usually about 48 H) the cells are certainly still CD4 or CD8 positive. How are you removing the cells? The company will not give you any info about the flasks because the coating process is a proprietary secret...
