We routinely use Hoechst in our laboratory. We published a fairly thorough investigation of the dye properties, Scatchard and Swartz plots comparing various fixations, concentrations etc. of Hoechst 33258 to 33342 (J. Histochem.Cytochem. 36:757-762). We now also know that the cells can also fixed in 0.5-2% paraformaldehyde (PF) for 15 minutes followed by 25%buffered ethanol with no effect on the binding curves (we only investigated Hoechst 33258 for this). Higher concentrations of PF introduce higher CV's and begin to distort other features of interest.
