Ken, If you sort beads and count after sorting you will get counts as you suggest routinely. Sort fixed cells into media with 20% FCS and you will find that your counts will be much closer. If with this test you still get counts as such then your drop delay is off or your abort rate is too high. Good Luck David Hi All, I'm seeing a discrepancy in what the Vantage sorter tells me it's sorted and the actual numbers sorted. For example, I sorted for green calibite beads (30,000) and when I re-ran them to check for purity there were roughly 15,000 only. Does anyone have the same problem?
