Dear Mark,

I am happy to inform you that we have been able to make the first and so far
only monoclonal antibodies against native human granzymes which can be used
in different immune assays.
For flow cytometry we made a PE-conjugated antibody against granzyme B, for
granzyme A you can use our biotin conjugated antibody.
So far most studies only focus on Granzyme B, although there is evidence
that granzyme-A can induce apoptosis through a delayed pathway or as
recently published it can even be depending on the target cell, Pardo J.,
Eur. J. Immunol.2002.32:1980-1985.

I can not help you with an antibody for perforin but this will be easier to
find. Please note that the function of perforin is now known to be involved
in the delivery of the actually apoptotic granzymes and thus less relevant
to study for CTL-activity, S. Metkar, Immunity, 16, 417, March 2002.

On our web site www.clb.nl <http://www.clb.nl> (go to reagentia) you can
find product information, protocols, example dot plots and a list of
references.

Also for these products we have free samples to test.

The latest news on the field of our granzyme antibodies is the development
of an ELISPOT kit, expected for release in November 2002.
It is evident that ELISPOT is getting more and more used since it is easier
to run many samples and much faster but comparable sensitive to flow.

To evaluate our kits we are looking for test sites, studies with relevant
patient groups etc. no ELISPOT experience required!

Best regards,

John

	List members,

I want to stain human CD8 lymphocytes for the expression of
perforin and granzymes A and B. I'd appreciate any references to currently
available antibodies for this purpose. Thanks in advance.
