activation markers

Dear experts in flow cytometry,

I have two questions regarding markers of activation during
persistent viral infection :
1. When performing functional assay in patients with chronic viral
infection, do all tetramer+ cells secreting INF/TNF after peptide
stimulation express activation markers like CD69/CD38 ? Which of
these activation markers do you find more appropriate in this case?
2. Does effector, viral specific PBMC CD8+ express activation
markers, or that would be only the case of CD8+ T lymphocytes that
migrated directly in the site/tissue of inflammation.
