Many years ago, back in the days when I wanted to get rid of them rather than study them, we found a very simple and highly effective way to get rid of platelets prior to separating mononuclear cells. We used "defibrination" as our anti-coagulant. This has the effect of avoiding the use of any other anticoagulant and totally depleting platelets (with the possible exception of some attached to monocytes). It has the downside of activating the coag cascade, which may or may not affect some leukocyte functions. If this technique is unfamiliar or of any interest I'd be happy to supply the "protocol".
