I have a client who is looking at ploidy levels in tomato plants. When we run her plant tissue through we get nice results. She has tried to stain some of her tissue culture cells and the histograms get pretty ugly. She was wondering if starch granules take up PI and if this might be why the nice clean peaks start looking like mounds. Any ideas on this one. I could not help with that question, but told her I would pass it on to the group. Thanks in advance for any and all input.
