An associate of mine is planning a rather ambitious experiment with a large number of samples to be analyzed with flow cytometry. He posed the question as to what effect if any, fixation of the cells, would have on fluorescence emission for the particular markers is interested in. He wants to stain the cells and then fix them, due to time constraints, and analyze them the following day. He is interested in measuring CD3, CD4, CD8a, CD11b and CD40 in splenic and lymph node cell populations. Does anyone have any experience or suggestions for the fixative of choice and whether analysis the day after fixation is reasonable. Thanks. Richard J.
